探討虱目魚精功能特性之抗發炎、抗氧化、提升免疫能力

國人在近十年來對養生保健的觀念越來越看重,也因此讓許多的養生保健的食品有了蓬勃發展的機會,因此開發養生產品滴虱目魚精、滴牛肉精來協助畜牧、水產業者創新產品並降低損耗的發生。本研究目的探討是針對產銷履歷虱目魚全魚滴煉成液體為主,全魚萃取完後的液體來檢測對人體有益能補充人體所需的蛋白質、胺基酸、牛磺酸、膠原蛋白、提升免疫力、抗發炎、抗氧化之功能特性,依牛肉精產品來做比對組。結果顯示高濃度之冷凍牛肉精(BF)、冷凍虱目魚精(FF)、常溫虱目魚精(F)可刺激Raw264.7細胞分泌細胞激素TNF-α、IL-10及IL-6,呈現劑量依賴性,且作用時間越長,細胞激素濃度分泌量越高。在抗發炎的部分,三個樣

品在作用4小時即可延緩LPS所誘導的TNF-α。而對TNF-α產生影響分析:其最適濃度在5%~2.5%之間。由此看出樣品刺激免疫反應可能偏向於Th1型,具有調節免疫潛力。實驗中發現體外抗氧化測定中,當產品濃度越高時對於抗氧化活性有顯著性的差異,攝取樣品原液100%對人體的抗氧化有一定程度的幫助,透過細胞試驗、體內外抗氧化試驗能得知傳統產業陶甕所生產出來的虱目魚精、牛肉精等等產品對於國人身體保健是有益處。關鍵字 : 虱目魚精、抗發炎、巨噬細胞、免疫調節、抗氧化、產銷履歷

扁實檸檬皮萃取物對大鼠藥物代謝系統及其抗氧化活性之影響

台灣香檬又名「扁實檸檬」,為台灣特有的柑橘類,含有豐富川陳皮素 (Nobiletin,NOB)及橘皮素(Tangeretin)等植物類黃酮,先前研究證實大鼠餵食扁實 檸檬乾燥粉具抗發炎、抗氧化、降低體脂及肝脂之機能性,而川陳皮素為扁實檸檬 皮中含量較高的機能性成分,因此本實驗想深入探討扁實檸檬皮萃取物(Citrus depressa Hayata peel extract, CDHPE) 對肝臟解毒代謝系統及抗氧化酵素活性之影 響。實驗分為三大部份,第一部份:以 5 隻 8 週齡雄性大鼠進行管餵 CDHPE 後, 採集血液及組織進行分析,大鼠管餵 CDHPE (含 50 mg/kg NOB)

後採集血液,測得 血中 NOB 濃度曲線下面積(area under the curve, AUC)為 1.43μg/ml × h、血中最高濃 度(Maxium Concentration, Cmax)為 0.52μg/ml、NOB 達血中濃度最高之時間(Tmax) 為 0.45 小時、平均滯留時間(Mean retention time, MRT)為 3.3 小時、半衰期(Half- life, T1/2)為 2.6 小時,組織分佈測得 NOB 及 Tangeretin 兩種類黃酮化合物,在胃中 顯示含量為最高,其次是肝臟。第二部份:將 18 隻 7 週齡雄性大鼠分成 3 組,分 別為:(1

)控制組(Control)、(2)低劑量組:1.1% CDHPE (50 mg/kg NOB,飼料中含 1.1% CDHPE)、(3)高劑量組:2.2% CDHPE (100 mg/kg NOB,飼料中含 2.2% CDHPE),連續處理 7 天,取血漿、肝臟及小腸組織進行分析。實驗結果顯示,在 小腸方面,CDHPE 會增加小腸 CYP1A1、CYP3A 活性,表示短期攝取可能會增加 phase I 藥物代謝能力。在肝臟方面,會增加 CYP1A1、1A2、2B、2C 及 2E1 活 性,並提高 UDP-glucuronosyltransferase(UGT) 及 glutathione S-t

ransferases(GST) 的 活性,降低活性氧(reactive oxygen species, ROS)含量,結果顯示 CDHPE 會提高肝 臟藥物代謝能力並降低肝臟氧化壓力。第三部份,將 10 隻 8 週齡雄性大鼠分成 2 組,分別為:(1)控制組(Control)、(2) 1.1% CDHPE (50 mg/kg NOB,飼料中含 1.1% CDHPE),連續處理 7 天,老鼠隔夜禁食後給予苯并[a]芘(Benzo[a]pyrene, B[a]P) 25 mg/kg,觀察 6 小時內 B[a]P 之血液濃度變化,結果顯示 CDHPE 會降低 6 小時內B[a]P 之血液濃度(AU

C) 、血中最高濃度(Cmax)及 B[a]P 達血中濃度最高之時間 (Tmax)。
